2) Dosage and usage 

As sildenafil, the recommended dose of 25 to 50 mg is administered orally once a day about 1 hour before sexual activity, and in some cases, it can be administered 4 hours to 30 minutes before sexual activity. The dosage may be increased or decreased depending on effectiveness and tolerability.

The following patients may increase the plasma concentration of sildenafil, so the initial dose should be adjusted to 25 mg.

- Age 65 or older (AUC 40% increase)

- Liver failure (cirrhosis: 80%)

- Severe renal failure (creatinine clearance less than 30 mL/min: 100%)

- When used in combination with CYP3A4 inhibitors (ketoconazole, itraconazole, erythromycin (182%), etc., saquinavir (210%)), the effect and adverse reaction rate may increase as the blood concentration increases.

- For patients taking ritonavir concomitantly, the maximum single dose in 48 hours should not exceed 25 mg.
